Body Burdens and Dietary Intake Elevated Dioxin Levels in Chloracne Cases Twenty Years after the Seveso, Italy Accident

چکیده
In July 1976, an industrial accident contaminated a residential area surrounding Seveso, Italy, with high levels of 2,3,7,8-Tetrachlorodibenzo-p-dioxin (TCDD). The exposure was acute, relatively pure, and affected more than 45,000 men, women, and children. By February 1978, 193 chloracne cases, mostly children, had been identified in the exposed population. 5 Twenty years after, we conducted a case-control study on subjects diagnosed with chloracne and control subjects, who had not developed chloracne after the accident, to evaluate their TCDD plasma levels, as well as the exposure-response relationship and possible determinants of susceptibility to TCDD effects in this population.

Tetrachlorodibenzo-p-dioxin (TCDD) is classified as a human carcinogen and exerts toxic effects on the skin (chloracne). Effects on reproductive, immunological, and endocrine systems have also been observed in animal models. TCDD acts through the aryl hydrocarbon receptor (AhR) pathway influencing largely unknown gene networks. An industrial accident in Seveso, Italy in 1976 exposed thousands o...
متن کاملCancer incidence in the population exposed to dioxin after the "Seveso accident": twenty years of follow-up
BACKGROUND The Seveso, Italy accident in 1976 caused the contamination of a large population by 2,3,7,8-tetrachlorodibenzo-p-dioxin (TCDD). Possible long-term effects have been examined through mortality and cancer incidence studies. We have updated the cancer incidence study which now covers the period 1977-96. METHODS The study population includes subjects resident at the time of the accide...
متن کاملAge- and Concentration-Dependent Elimination Half-Life of 2,3,7,8-Tetrachlorodibenzo-p-dioxin in Seveso Children
OBJECTIVE Pharmacokinetic and statistical analyses are reported to elucidate key variables affecting 2,3,7,8-tetrachlorodibenzo-p-dioxin (TCDD) elimination in children and adolescents. DESIGN We used blood concentrations to calculate TCDD elimination half-life. Variables examined by statistical analysis include age, latency from exposure, sex, TCDD concentration and quantity in the body, seve...
متن کاملUrinary D-glucaric acid excretion in the Seveso area, polluted by tetrachloro-dibenzo-p-dioxin (TCDD): five years of experience.
On July 10, 1976, an explosion in a factory in Seveso, Italy, located 30 km north of Milan, producing trichlorophenol caused the release of TCDD-containing compounds in the surrounding area. Since extremely small doses of TCDD have been shown to induce hepatic microsomal enzymes in animals, urinary D-glucaric acid excretion (a measurable index of enzyme induction), has been investigated in Seve...
متن کاملShort- and long-term morbidity and mortality in the population exposed to dioxin after the "Seveso accident".
The early effects of 2,3,7,8-tetrachlorodibenzo-para-dioxin (TCDD) exposure in the population involved in the Seveso, Italy, incident in 1976, have been examined in numerous studies. Chloracne was the only effect linked with sufficient certainty to dioxin exposure. The possible long-term consequences were investigated with mortality and cancer incidence studies. Mortality and morbidity findings...
